How It Works

Selank is a synthetic heptapeptide derived from the naturally occurring immunomodulatory peptide tuftsin, with an added Pro-Gly-Pro sequence for metabolic stability. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, it has been approved in Russia as an anxiolytic and nootropic since the early 2000s.

The peptide's primary mechanism involves modulation of GABAergic neurotransmission. Unlike benzodiazepines, Selank does not directly bind GABA-A receptors. Instead, it alters the expression of genes involved in GABA synthesis and transport, producing anxiolytic effects without sedation, tolerance, or withdrawal. Selank also stabilizes enkephalins — endogenous opioid peptides involved in mood regulation — by inhibiting their enzymatic degradation.

Additionally, Selank influences serotonin metabolism and has demonstrated immunomodulatory properties, including effects on IL-6 expression and other inflammatory cytokines.

Benefits

  • Significant anxiolytic effects — comparable to benzodiazepines in animal studies without sedation or addiction risk
  • Cognitive enhancement — improved memory and attention under stress conditions
  • Immunomodulatory properties — regulation of inflammatory cytokine expression
  • No tolerance or dependency — safe for repeated use without withdrawal
  • Mood stabilization — through enkephalin and serotonin system modulation

Side Effects

  • Exceptionally well-tolerated in clinical use with minimal reported side effects
  • Nasal irritation — common with intranasal administration
  • Mild fatigue — occasionally reported, particularly at higher doses
  • Headache — rare
  • No sedation, cognitive impairment, or withdrawal effects documented
